Do I need to do all of them? Do they keep spawning or will they eventually end?

This will definitely take more than 100hs to complete 100%

Another thing, is the NG+ worth it? (Aiming for the other endings and achievements)

The NCPD gigs are all placed there, and they do not respawn. There are achievements for completing them and all the fixer-gigs in each of the city districts.

You can see them on your map using the custom-filter so you can plan to do them in between other things.

The game does not have NG+, but I think there are mods for it.

To get all the achievements in one safe, you need to save before the final mission and go through each of the proposed endings. At least it's better than replaying the game every time dude...
