Depending on how far your save is in, may miss some early added iconics like the new knife from the Heist. But thats only major thing can think of

There are certain main game side quests that add options to Phantom Liberty side quests/gigs but besides that PL is its own thing. It does add a new game end path.

Also yes no spoilers. Dont read any.

No it doesn't. Its a Mid game DLC in a separate region of the city. It is not a expansion or sequel.

Idk, from what I read it can technically replace act 3, though also people are saying don't start it until the point of no return in act 3.

No especially if you already started a new game in the 2.0 update.

There nothing you can't discover later on in a new play through. I continued my previous save for both 2.0 and PL

PL is whole new area to explore. You can even chose to skip the DLC by not performing certain actions. Yes there is a new ending

(1068 hrs on GOG)

I started a new game to get to Phantom Liberty, and my only regret is that I didn't play more of the base game before hitting Dogtown. But then I find this game very, very repeatable (different genders, different Lifepaths, different romances, different playstyles).

As far as I can it's not that was important before I pulled the trigger with an old character and went into PL before Nocturne OP. I'll tell you this a new character will have a hard time leveling up[ without the advantage of all the old XP gained through missions before the non DLC Nocturne Op and side jobs and NCPD scanners. I just respecced my old and dove in.
