No, you get to choose which ending you want between the DLC and the main one. Once you completed all the DLC main quests, you will need to call a certain someone to go with the DLC ending. But even if you call them, you can delay the ending to go with the main game ending.
4 for the DLC questline that are "midgame" endings with their own credit scroll and you get put back into the world to continue the rest of the base game as normal.
the DLC also adds 1 (depending on choices made) additional ending that you can select to go down when talking to johnny on the rooftop.
You can start and finish the DLC and it doesn't lock you out of anything, during or after. You can come and go from the new area as much as you like.
There is a "secret" new main game ending that you can unlock toward the end of the DLC missions, and if you unlock it, you'll get a point-of-no-return mission after you finish the DLC that you can choose to start or not. If you do begin that mission, then yes, you would be essentially finishing the game. If you don't do that mission, you can continue to do what you like, even ignoring it completely for the main game endings.
