Next update (in the next few months?) Introduces NPC animals. After that human NPC's should make their way into the game. Story will likely be last if I were to guess.
Once animals get released you can expect many new mods on the workshop, including better NPC mods.
The game has been striving for realistic depiction of zombie apocalypse. As realistic a zombie apocalypse can be, at least. So there probably won't be any grand quests where you save the day by finding the cure and everyone clapped, and whatnot. Probably some minor stories like what's already seen in game through house and road events at most.
I do hope we would have at least some raids happening a la Horde Night mod.
Asides from that I don't think so. Maybe NPCs will patrol the fence to Louisville to keep you out and then are overrun once the zombies break the fence in the timeline along with things like that
