It's good to keep this part in mind, and it's why the downtown area is so dangerous.
Assume each building is likely housing >50 zombies. If you get caught in the middle of the street between two buildings, it's quite easy for zombies to flood in from the front and the back making both forward progress and retreat impossible for vehicles. By the time you're properly surrounded and unable to move your vehicle, it's also too dangerous to leave it and try running.
It's much better to start blasting your weaponry from areas you KNOW there will be fewer/no zombies attacking. There are places like the mansions along the river or where Bender mentioned in the Northwest corner of Louisville. This will allow you to always have somewhere safe to run/drive back to.
Also, when you get a smaller vehicle, it helps to keep in mind that you can perform drive-by shootings with your guns (or even melee attacks, but I don't recommend it due to zombies also being able to attack you). You can drive down to an area, shoot a few shotgun shells off, then tear off down the road a few blocks. Many zombies will pour into that area and out of the surroundings making them easier to loot.
Another thing I don't recommend is using fire. There's a lot of good stuff in Louisville, using fire has a chance of spreading to buildings you don't want razed. It's densely packed so fires have a tendency to spread everywhere.
Yes. You must roll the window down first, though. This is what allows zombies to attack you back. To avoid this, continue moving (either forward or reverse) and their attack animations will not land.
There's a cruise control function for vehicles. You can set this to a low speed so you only need to worry about aiming and steering. I use a controller and have been unable to set it with a controller, however the keyboard commands work.
No, it wouldn't make sense. At most, you can attack 3 of the zombies with vanilla weapons. 2 with a baseball bat. If you open the window, you will immediately be attacked if already surrounded. Those attacks will block you from attacking back. Best case scenario if fully surrounded is to power through ahead or reverse until you're clear enough to get out and run. It's tough.
They can't pull you out of the vehicle, but they don't have to. One bite is enough to end you and the odds of getting one (or scratched/lacerated) are high when fully surrounded.
I can't see fighting from a big truck in streets clogged with zombies ending well. Best bet is to not get surrounded. And that means find a compact to go scouting in. I do hope you'll post how you get on. feeling quite attached to your adventure now :)
The long journey is the funniest thing we did with my brother, ♥♥♥♥ happened, most supplies had to be abandoned but eventually we made it. Without picking a spot we drove there and ended up with a whole square as "base".
My suggestion is to drive into Louisville North East Center all at once, spotting police cars and ambulances. Those are needed to hoard entire buildings. There are a lot of fenced areas (werehouses) near public buildings that work as provisory bases. I believe you can clear out that area first to safely move west to your luxury home.
Also, if you want tips where to settle in there I'd suggest this house

It has almost complete high fence around it and a body of water in the backyard. There's a gas station nearby too. Just know that there will be more zombies in Louisville than anywhere else on the map :) At least that was my experience! I visited some mall there too and there were so many zombies that they were falling down from the floor above on top of you. That was pretty intense but extremely fun to see. There's army surplus store in the northern part of the city if you need some army clothes and other stuff. If you have Brita's weapon pack it's time to bring the minigun with you...
