How to restart the floor with no repercussions
By The Source of Your Problems
Disclaimer Alert: THIS IS CHEATING, if you like playing legitimately, then in all likelihood this guide is not for you.
It is possible to go through a floor normally, find item room/shop/boss etc, then reset the floor. The timer will go back, Items will go back, and you can change the path you take to get wherever you want.

Why you would want to do this
Have you ever been on good run, going for boss rush, but you just barely missed it because you didn't know the floor layout? Or maybe you pick up an item that causes you to repeatedly damage yourself to the point where it makes the run unplayable? Well, if you keep reading, you will learn how to go back to the start of the floor, back to a point where you hadn't picked up the bad items, and the floor layout doesn't change a bit!
How it works
TBoI saves your progress on a run at the start of a new floor, whenever you leave the game (pressing pause, then exit game), or whenever you close the game (through alt+f4 or equivalent).
However, the game does not save when it crashes. We will be using this to our advantage.
How to crash your game
1: Open task manager. You should already know how to do this, but if not, press Ctrl+Shift+Esc.
2: Click on "Performance" at the top left. This will show you your computers performance, but whatever we aren't here for that.

3: Click "Open Resource Monitor" at the bottom left. As it says, this will open the resource monitor.

4: Find isaac-ng.exe, right click on it, and click "Suspend Process".

5: Wait a few seconds, then right click on isaac-ng.exe again, and click "End Process".

If you followed these steps, the game will have crashed, so reopen it. Then when you press continue, you will have been taken back to either the start of the floor, or the last place you exited the game at, depending on which happened latest.
Enjoy :)
Congratulations, you cheated. Whatever reason you may have to do it, just remember that nobody really cares if you do or not, because this really only works in a single player game.
Well anyways, thank you for reading, and I hope you find rock bottom in your next secret room!

Or you can finish the floor and in the short animation between floors press Alt+F4

I just tested it with Repentance (mods disabled) and it worked fine for me. I may have neglected to mention that you can only do this while still alive, as I believe that dying saves the game as well. Otherwise, let the game stay suspended for a few more seconds (minimum of 7 seems to work every time). Let me know if it works for you then!

i think it only works for before repentance

At least one of the DLCs prevents this from working
