What is this I don't even???!!!1
I presume you wish to know what these rooms are. Well, Angel Rooms are little gems that have a chance of appearing at the end of a boss. Like Devil Rooms, these will never appear on Floor 1, but they may appear on any floor after that (apart from Floors 9 and 10).
At first, the chances of one appearing are slim, but there are ways to increase these chances (see below).

Once you're in the room, first notice the angel in the center. He presents you a randomly selected item from a selected collection (see below).
How do I get in?
Right. This is a tricky question, because a lot of the following methods of increasing chances for one to appear are not confirmed and the pecentages are unknown. However, Edmund McMillen has confirmed that you can by:
  • Taking no red heart damage during the boss on the current floor
  • Owning an item that gives "Faith Up"
  • Giving to Beggars (both types)
  • Having previously entered an Angel Room.
  • Refusing to take items from a Devil Room.
The following are unconfirmed:
  • Using The Bible a lot.
  • Using Blood Donation Machines.

What appears in there?
This is a list of the items you can get in an Angel Room. Note that these can be rerolled by the Dice into another item from this list.
(IMPORTANT: If an Angel Room appears after killing Mom's Heart, it will contain a trap door to Floor 9 instead of an item.)

The Bible
Useable Item; Space to use (Cooldown 6 rooms)
Gives Isaac the ability to fly over obstacles for 1 room. Kills Mom and Mom's Heart instantly during their fights, but if you use it on any other boss after those, it will deal 12 full hearts of damage to you.

Dead Dove
Treasure; Passive
Gives Isaac spectral tears and the ability to fly permenantly.

The Halo
Treasure; Stat Increase
Increases all stats except Range.

Blood of the Martyr
Treasure; Stat Increase
Increases Damage by 2 and gives Isaac blood tears.

Guardian Angel
Treasure; Familiar
Flies around Isaac, blocking projectiles and causing damage on touch. Doubles the speed of all orbiting items.

Holy Grail
Treasure; Stat Increase/Passive
Gives Isaac the ability to fly permenantly and 1 empty heart container.

Prayer Card
Useable Item; Space to use (Cooldown 6 rooms)
Gives Isaac an Eternal Heart, or a container if he already has an Eternal Heart.

The Relic
Treasure; Familiar
Follows Isaac from room to room, dropping a Soul Heart every 4 rooms that are cleared.

Sacred Heart
Treasure; Stat Increase/Passive
Restores health fully and gives 1 Soul Heart on pick up. Shot size and damage is greatly increased, and gives homing tears. Shot speed and Tears are decreased.

Scapular
Treasure; Passive
When Isaac has only half a heart left, this will grant him 1 Soul Heart, giving him a last chance.

Mitre
Treasure; Passive
Increases the chances of getting Soul Hearts in place of normal Red Hearts.
Notes
From the BOI Wiki:

The reason the angel is missing a face is likely a reference to the Bible passage Exodus 33:20: "But,” he said, “you cannot see my face, for man shall not see me and live.”

If I missed anything, be sure to say so in the comments. I hope this guide helped you.

Also, if you go to a super secret room which coincidentally has the design of an angel room then use any item dropping items. Such as lovers which gives to 2 half eternal or spirit hearts. if you have the portable slot machine then all hearts dropped will be eternal hearts. I tried it once and I was surprsied.

No Mitre? You also forgot the fact that a Joker card could TP you in one.

Elucidate, faith up was supposed to be a factor in multiple things, also leading certain characters to Angel Rooms or Devil Rooms. Certain items were supposed to somewhat correspond to faith, but in the end, they were just lazy about it and forgot about it.

Just got into one by opening a Red Chest through a fanged door.

Hey, quite a few things wrong with this guide:

"Owning an item that gives Faith Up" - This is a common misconception where no actual proof has been discovered that "faith up" items give higher chance for an angel room. If you can provide evidence then great.

"Giving to Beggars (both types)" - False, you get +35% if you kill a beggar of any type on the current floor.

"Having previously entered an Angel Room." - Again not sure about this being true, there's nothing about this I can find in the game's source code.

"The Halo - Increases all stats except Range." - False again, the halo does give range up. The wiki is wrong about this and so many other things, use multiple sources.
