The relics they have (ribcage and heart, as said) are not so important, in my opinion, because another "successor" has the "breath" that has a symbol depicting the lungs, which is very close to both, but the person is completely unrelated. Aurora's memories, the way she is first against the whole plan, then agrees with complete conviction explicitly to help Karen, reinforce how important she is to Aurora, and suggest that Aurora would've accepted any of the relics as long as she can do anything in her power for Karen and stay by her side.
Another detail worth mentioning is that most of the successors (relic keepers) are located very far from each other, because the whole point is to separate the relics. As far as shown in the game, Aurora and Karen are the only "entombed" ones so close, one protecting the other, even if Karen has a unique role.
Every "final" interaction with Aurora is about Karen only, too, and in the good/true ending they keep working/staying together.
Despite all this indirect evidence, though, it's not clearly indicated that Aurora's feelings are romantic, or whether they would be reciprocated if they were.
As for Io, while her mission from the start is to be completely devoted to the MC's well-being, it's way easier to interpret it as not having to do anything with romance. (The MC's default gender, based on the intro that can be watched by waiting at the main menu without doing anything for a minute or so, is male, too, so I don't mind that there isn't any dialogue option to make the relationship with her sound romantic.) I'd rather summarize it as the relationship turning into an equally trusting and loving friendship in the best case. In a sequel, based on the true ending, there could be potential for something more with Io, but the game's creators typically don't push romance between the MC and any NPC openly, as far as I saw in their games.
After all these spoilers, the most I could think of adding would be "It's strongly hinted at that a female NPC has romantic feelings for another female NPC." Since it's not completely confirmed, that may rightfully not be enough for the game to be listed. (Even if that'll always be my interpretation personally, including that it's mutual, unless the opposite gets confirmed.)
Io's relationship with the MC pretty much falls under the same category as these relationships. And yeah, it's definitely because they've always defaulted with male protagonist with all of these games they've made. It's why the mc is more supportive with Mia when you have to delve into the memories of the Breath, holding her hand and guiding her more closely compared to every other successor, cause Mia also falls under the same heroine role that Alisa, Ciel, and Claire did in the GE games. The Ribcage and Heart imagery has significance when paired up with other factors like where Aurora and Karen are situated- right next to each other with Aurora guarding Karen as a giant dog. I'm very much doubtful that they just randomly picked ribcage to be the one protecting the successor of the heart. The breath doesn't matter there cause it has no other elements supporting it.
