If someone in a Batman suit walks up to you, right now. You will think: "Hey that's Batman!" How is that not real?
Anyway, I made an attempt at your top 5, but it was impossible in such a short timespan to make any proper list.
I already got wile e coyote, because it's a simple concept, and as viewer you're sort of hoping he catches the roadrunner, because the roadrunner always gets away, and you never know where it´s going, so that makes it a likeable villain, and he also looks hungry, and fate punishes him always with his own delivered ACME contraptions, yet he always survives, and that´s just perfect, because it´s funny. And you sort of like him, because of it. It´s like justice being served right there, by his own doing.
I´ve got the black witch, with the poisonous fruit and the mirror, which is also a well developed villain.
Whatever is being used for an exorcist, also makes for good villain material. That´s when the villain knows every detail about someone. Except the exorcist is also a villain, so you have two, but you tend to favorite the one in chains.

Jaws. Because it's funny to have a fish with a grudge. It's also possible, fish can be quite mad. But I'm not sure if you would consider that creature fictional or not. But it should still get on your list, for the sake of having some kind of animal there.
