Mother board is Asus P6T. Need USB 3.0 20pin for USB 3.0 front I/O.
What are my options?

Those are the specs. I am only using a graphics card for one pci-e x16.
I'm buying a new case that will have USB 3.0 for front I/O.

I see that there are pci-e adapter for usb 3.0. In real-time, will I see a difference between using pci-e 1.0 vs pci-e 2.0 ?

USB 3.0 isnt really all that important, while it does have better transfer speeds compared to USB 2.0 you'll only notice the difference with USB 3.0 capable devices, such as thumb drives and external USB hard drives.

USB and PCI/PCI-E are not the same.

I'm going to be buying a portable hard drive. I will use it on that. Do the portable hard drives like the really slim WD my passport use disks? just curious.

if your case has usb3 ports with the usb usb3 card ChrisW linked

if the case doesnt have a usb3 port, get a e-sata port for it, and an external drive with esata

How big of a HD are you looking for? You could just go with a 32GB-64GB USB Thumb drive which would cost quite a bit less in many cases compared to an external HD and may have the same performance.
