Nvidia App Auto-Tuning
Does it make sense to activate this setting as a normal gaming user, or can it be safely ignored?

Always used MSI Afterburner when using Windows, so I'd vote for ignore.

if you do not know how to change game settings

sure let it change them for you

but if you can go to visual settings in game, or modify config files dont

This setting is not about the graphics settings of a game, but about overclocking the GPU. Something I have never dealt with.

the new nvidia app (previously gfe) also has settings to change game settings for you for 'optimal fps' whatever they think you should get with your cpu/gpu

gpu overclocking is also kinda pointless, they already boost to whatever they can do as long as they stay cool
forcing a higher oc will make them reach max temp sooner and throttle sooner and lower to keep cool
