Does it matter in which ram slot a ram stick goes in
So Im about to buy aa 8GB Kingston HyperX fury, but here's the problem though. the way the ram stick is built will probably hit CPU heatsink cooler, but the other 2 slots aren't slightly blocked. So would insterting the stick in the 2nd slot have undesired effects?

Read your motherboard manual. It will tell you which slots to insert ram. It all depends on the motherboard and the type of channel.

For example, if your motherboard is dual channel and you use a single stick or put 2 sticks in the wrong slots, you will not get the performance benefit of dual channel.

With 2 sticks it's every other slot. If your heatsink is blocking it then try sliding the fan up if it's detatchable.

Feel free to insert DIMM in any available slot. If it will be your 2nd DIMM, you, in worst case, will stay with the same single-channel RAM which you already have. Really, for gaming purposes with discrete graphics, difference in single- and dual-channel FPS is negligible.

fill the same colored slots first

If one only uses their PC for gaming, then single channel will definitely be just fine.

If one uses their PC for more than gaming, then the difference in single/dual/triple/quad is worth getting a better CPU cooler with appropriate RAM clearance or get RAM that has a shorter heat spreader.

for amd apus the ram speed makes a direct difference on the gpus performance
but thats easily outclassed by a decent cpu and dedicated gpu

single/dual ch for most make very little difference
but if he has 2 or 4 similar dimms, its best to run them in dual ch modes
Yes it does, if you want multi-channel bandwidth. You want to run in pairs for dual, triplets for tri channel and fours for quad channel, each dimm slot is colour coded for the channel grouping.
