Green Download icon to the right
Open Shutup10
Second section, make sure Windows Defender is Green, if it isnt it isnt fully disabled, so click it.
Under Actions
Apply All Recommended Settings, i recommend doing that.
You may also go through the list and see what options there are and what can be disabled.

Reboot when finished
You can also use Shutup10 to Re enable Win Defender, reboot, then disable it via Shutup10.
Just to make sure
and tick everything in the OS back ON again, so OS Defaults.
Now reboot to apply. Then re-run the app again and setup how you want once again, then reboot to apply.
Please extract Shutup10 to a folder you will always run it from, before you ever run it, cause it will output an INI file which must reside in the same folder as the app's EXE file.
Do a full scan with Malwarebytes 3.x
