Can i use a CPU-OPT header for a regular case fan ?

Yes of course.

The reason many higher end boards have those; is to account for users who might use an Air Cooler with more than one fan; or Liquid Cooling for example with Rad + Multiple Fans you like to have read as "CPU related fans"

Overall it really doesn't matter; as long as actual CPU Fan is on the main / primary CPU fan header for obvious reasons. As you want the proper alert and such if there is a real issue with actual CPU fan.
