It might be incompatible with that motherboard.
Which is the models for Motherboard and RAM?
What OS are you using?
Did you verify the ram upon boot up, in the BIOS?
Make sure that all the RAM you use is same Mhz, CL and V.
It would have been better to get another 2x2 Gb with same Mhz, CL and V values if you cant get exact match for brand and model.
RAM is best to be installed by matching pairs so you dont lose the dual-channel effect.
Just one stick of 4 Gb should work though without dual-channel effect, could be just faulty stick.

Asus P5QL-EM
4x DIMM slots @ 8GB Max
2GB per DIMM slot

If want to do 1066Mhz, only 2 (yellow) slots can be populated

it is a good idea to always update your bios when doing an upgrade, if there are any.

put it on a usb and boot from the usb.

:qr:

Do I download everything or just one?

the most recent bios update was long ago. you may not need to update it

:qr:
Just the latest bios version; it will contain the fixes from all the rest as well.
The download will be a compressed file; extract the contents to usb flash drive.
Then can update it from within the bios and it'll read the file from the flash drive.
Did you try that 4GB one by itself?
According to the motherboard specs, it does not appear to support any 4GB as a single dimm.
8GB total divided x4 = 2GB max per dimm

I thought I had made that clear, as it only took looking up the motherboard model to figure this out, and quickly. Now in the support page it does say some were tested via "4GB (kit of 2)" but this means that those were a matching 2x2GB Kit

Could you please listed all the DDR2 you have available to try and their models?
Then we could best determine which can all work together well for your board before you return/buy anymore RAM.
