Did you try remove the WiFi card so you can then test the Capture Card in that slot, so ensure the Capture Card actually does fully work?

Motherboard have a limited amount of what all the Chipset can handle; USB / SATA / PCIE-Lanes; etc. And slots usually share with something else on the board/chipset.
So if you are using a Graphics Card in PCIE X16 Slot #1; one of those PCIE X1 might not function when that is populated.
Again I would (since easy to do) remove the the WiFi Card from PCIE X1 (physically slot #3) and try the Capture Card on that. As it could be you can't use PCIE X1 (slot #2) at the same time; or it could be a faulty card.
If the Capture Card does work, but you can't use it in PCIE X1 #2; then the easiest route here is ditch the PCIE WiFi card and go with one that is USB based. Or perhaps buy a Capture Device that is USB 3.0
Since as I understood the problem is that graphics card heatsink will just cover the PCI x1 slot next to it so you cant physically instert card in there.

But yea if 3x PCIE slots total and using a GPU, you can rule out using slot#2
I don't even think they can be found in USB 2.0 anymore; maybe back when 720p was the norm.
