Remove it from RAMDisk, update the game with it stored on local disk.
Once update has complete, verify the game cache in Steam.
Then reinject it into RAMDisk once again.

RAMDisk is separate temp injection; remove that from RAMDisk.
Update the game and then re-inject it; very simple.
Whats inside RAMDisk is not real; it is read-only.

no, it's not read only

The update SW may require double space, as it may want to get all the new files before deleting the old ones. Also the DL sice is not necessary the same as the install size, the DL may be compressed.

If you DL is fast and free easiest is to just delete the old files, and get them redownloaded.

If not, then I'd keep the game install on HDD and use a mirror of it on the ramdisk:
- install on HDD
- find the game directory (library_root\steamapps\common\<game>)
- copy that to ramdisk
- rename the original folder
- create a link with the original name that points to the ramdisk folder

when update is due, you can just repoint the link to the original
