If not then turn the on switch on (I've forgotten to do this a few times). There might even be a volume dial on the mic itself.
Also make sure you plug the mic jack(not the sound jack) into the mic port(not the sound port).
Beyond that, make sure your sound card/chipset drivers are installed properly and are up to date. Mess about with the Window's volume controls a bit until it shows activity when you're talking and near nothing when you aren't (adjust so your mic doesn't pick up your Hard-drives or case fans spinning)
Can you verifiy it works in steam chatrooms? (you and at least one other person have to dig for the menu to turn voice chat on).
If you can find any one person who can hear you in any other game (like TF2) then you have a game related setting that isn't picking up your mic. However there could be a few other things beyond your control. You may have been accidently muted by individual players or the server admin (or a script) decided to mute you across the board on their one server.
In summary, prove that the hardware works, then that steam itself is "hearing" you, then steam chatrooms(you can skip this if you want), another voice-chat enabled game, and finally the game you're having trouble with. If you still have trouble feel free to put your findings here.

Minor Note: Some headsets (low end products) that are wireless often have issues with co op games due to delay, recommend all devices to be wired; those milliseconds count ;D
