Does it matter if I install a game (Dota 2) not in the main directory (C:) but in D: or E: ?
I think my Dota 2 is lagging because of this?

It shouldn't matter. Right now (until I get my SSD next week) I run all my games off the second hard drive and I have no issues. My main hard drive is 500GB WD Black Caviar. My second hard drive is a 1TB WD Black Caviar.

The only problem you may have is if the hard drive is slow (5400rpm), then you may notice longer load times. Once a game loads, the computer is usually dependent on the VRAM in the video card and system RAM to help store/load images and the HDD should be used very little. If you lack enough VRAM and system RAM, then the computer will utilize the HDD (slowest part of the computer) to store information and this will cause noticeable slow down times. So, instead of the CPU being able to grab things from the RAM really fast, it needs to wait for the HDD to spin, locate and pull data from it.

It won't matter where the game gets installed to. However if that other drive is slow, then that can effect game loading, prefetching and overall performance to certain extent.

But for game like DOTA2 which doesn't require much to run well, I don't see it being an issue.

I know this thread ended a while ago, but I can tell you that YES, it does make a difference if the game is installed on a different hard drive or SSD to the one you run Windows on. I found that my game would run at 400 ping for as long as I had it on my (D:) drive and not on my (C:). It would only run normally when moved back on to my (C:) which was at about 25-30 ping.
