Nothing should involve a telephone call. I've received pre-paid AmEx cards in the past and used them on Steam. The registration is done entirely on the website. If you're picking up a phone, you're doing it wrong. OP and I were talking about two different kinds of cards.

Then you can register online and without a telephone call. I can't tell you the exact URL since I don't have an AmEx gift card handy but am 100% sure the URL is on the back of the card.
You first login with the card and the CVV. That gets you onto their website. From there, you should find a "Register Your Card" button somewhere that allows you to specify your name and mailing address.
It goes into effect immediately and you can purchase from the Steam Store immediately after registering. OP and I were talking about two different kinds of cards.

Note: You cannot register your card?s billing address online. You have to call AMEX Gift Card Customer Service at 1.877.AXP.GIFT to register an address to the card.

Give 'em a call. They are 24x7. Well, some portions of American Express are 24x7. Who knows with this card.
