I have top up and had purchased a lot of in game items (dota 2) but my Points does not increase even though i tried multiple times! Please someone enlighten me regarding this matter.

How did you get the money? Did you charge it yourself or did someone else send you a digital gift card?
The latter does not give Steam Points since the money was spend by someone else, that user will get the points.

Are you purchasing these items in-game itself or through the market? If it's the market then that doesn't count.

Market doesn't count - the purchase has to be made directly in-game or through the Steam Store to earn points.
