There are some Cyber Elves that you can only gather from mission-specific enemies, yes. It's a matter of killing a certain number of enemy in that given Mission.
Some elves aren't so stringent (you *can* go back and get them), but a number of them are only unlockable by killing enemies during the mission--if you beat the mission without satisfying the kill-count, you don't get the Cyber Elf.
Also: some Elves only appear in sections of the map available only during the mission! So explore every nook and cranny of the stage--but don't take too long or you'll get docked points on the mission rank!
That's only true for Zero 1 and for the elves that come from enemies, the ones in boxes can be collected any time you can get into the area. If you plan on getting all of the elves in one shot i recommend a walkthrough
It's not a challenge, it's the basic difficulty. In mega man Z2 if you want to earn all EX skills you need to be rank A so if you use elfs..........
If you want to unlock ultimate mode that can count too because you need to have all elfs in Z1 and Z2 (I forgot about Z3 and Z4)
