1. Yes, you can escort arrestees to holding cells yourself. You need to unlock it by collecting duty stars but it is a feature in the game.
2. Yes, you escort the criminals from your car to the holding cells and place them in custody.
3. No, the only multiplayer option is online.
4. Not a lot of these scenarios are in place yet, but I hear the devs are working on them. So far there are the typical duties (like parking tickets), traffic stops, accidents, drug dealing, graffiti, and crime scenes. You mentioned burglaries, and that kind of corresponds to the robbery crime scenes, which usually happen at shops or newspaper stands.
5. Yes, you will need to call EMT's at aggravated assault crime scenes and occasionally at accidents. Sometimes the pathfinding is off though. :(
6. Regarding the crime scenes, all you do is interview witnesses and gather evidence that way. It's a little unrealistic because ~75% of the time a witness tells you exactly who committed the crime. Other times they'll give you clues about what they were wearing or gender and such.

4. There's a dozen or so different interactions (i think) which includes the types of traffic stops. Callouts are unlocked as you progress through the game. So far I don't believe murder, arson, domestic violence are included.
