I killed one of the harpies to make things easier, and the others two say that they will not help me because i killed her sister, i will miss something?

You will miss a piece of equipment for Oro I believe. Spare all of them to unlock the next step of a quest.

You meet missing Izusa and Oro's final weapon, beating all 3 without killing is extremely difficult, I've tried doing it off and on for a while now with no success

Use freeze/petrify to temporarily remove one of the sisters from the fight. Also clear the adds to avoid combos and buffs.

Remember if needed you can down them (without finishing them off) and it will at least waste a turn of one of the sisters to go heal them.
