places like the cave, spider lair and other places can somehow be fulfilled so that they are not active on the map or are they always renewed later? Or can they be completed when I complete the combo? Thanks

They pretty much serve no purpose as far as I'm aware, they're just a marker in case you need to finish the 'defeat x amount of spiders' challenges.

so are they always active on the map? in another game, when something is selected or fulfilled, that place is no longer active

Yes, they're always active on the map.
I cleared out a few and they remained on the map, I think it really is just a spawning point for them.

From the few hours I've played only bandit camps can get completed and get a checkmark on them
