Can you use the character customization in the story campaign?

Reach is the only game that does this out of the box. There are mods for the other games that let you do this but it wasn't supported in the original versions.

Halo: Reach is the only game where character customization affects how you look in the campaign. I think some recent updates have allowed gun skins and vehicle skins to work in the Halo 3 campaign, but that's not a part of the original game.

Both of these work solo and in co-op.

It makes sense in the games where it doesn't allow for it. In Halo CE->4 you play as the Master Chief who has a set appearance. In ODST you play as multiple different characters throughout it's story.

With Reach you play as Noble 6 who's a more ambiguous character in terms of gender/armor style so the game lets you customise them.
