You need to go back to your previous autosave at which point she was still on Overbrook in order to make her reach ecstatic mood.
To go back to your previous save, there is an option on the screen when you're selecting the file you want to play.
To make Daria reach ecstatic mood, simply give her her favorite food: an apple, and give her a hug. These two actions will suffice. The mood indicating dot on the gauge will not go the the farthest right. But the ecstatic mood square box will be glowing, which means she is now at her max happiness.

Do I need to keep playing that save or can I reload, make her Ecstatic, then load my current save?

I don't know for sure, but my guess is that you need to keep playing until everyone has been ecstatic in the same timeline. Once you have the achievement, you can definitely revert safely, but I suspect whether or not a given character has been ecstatic is only stored in the current save file.
