what is the "Improved Item Quality" rune doing ?

It works like the toilet aura. It increases the attributes of items. They are based on your level, your toilet aura and now also on these runes.

I sacrifice first rune, and then start to gain money to find next rune, and when it apper the button to sacrifice one more rune, I received nothing. My progress was gone, but I still have 1 rune, it is a bug?

It is a rounding issue. You might have 0.995 runes but 1 is displayed...as the rune hasn't been completed yet, nothing will be added.

Collecting more runes is recommended anyway as 1 more rune won't make a difference. For example, collect runes for 24 hours, then sacrifice them.

I'd say so. You need to sacrifice them often as the more you have, the faster you will generated new ones.
