They really do yes, Persia and maybe the Armenians or Georgians (as most of the other factions were either under ottoman rule or already represented by the indians in the game)
I was thinking mamelukes or Sikh Empire but I don't know if they were independant, also it has to be a combo of a 2 nations from the same area ( you can't add Persia and Brazil in the same pack for example)

For consulate yes these civs you mentionned could be added but like Japan, choosing a "Native option" like "Iranic Isolationist" which would allow to recruit and benefits from native iranic units and features, example "Tajik Reinforcements" or "Afghan militiamen"
Yes, new natives and mercenaries could be added and be added to the Persian roster as cards some cards will allow you to train unique factional mercs and natives
Safavid dynasty as one of "gunpowder empire" would fit perfectly into AoE3
