As for the plague, I haven't beaten it yet either, just wait until the very late game before unleashing it and they'll probably be able to beat it.
Regarding the plague, I have a theory: Its nowhere written down, but I think you need a Snake for it. Snakes drop Venom and the flavour text of it reads like "is needed for medicine", so I guess you need medicine, adv. medicine as well as some venom to find a cure. You get a Snake when you play a critter card on a jungle tile.
i have beaten the plague a good few times now, and a general rule i personally keep, due to so many failures late game...
try your best to get the plague on an early bum rush...
the less people means you have less people to reinfect your "cured"
i had a world with close to 30 people on a small island... jill was cured 7 times...

the first time i managed to beat it, i spawned in a snake nearly the first critter, killed it with a card, and then "sampled" it a couple of times.
less people increases your odds of the first guy to get away from the buggers before they reinfect him/her :p
and yeah, it requires coraal, herbs, and venom, all researched (venom doesn't need research, but the others do) to finally beat the plague)
