What the paragraph in question says is this:
Player characters added by DLCs do not need to be unlocked and can be used from the start.
However, story dungeons first need to be cleared with their main character (e.g., Marisa's story needs to be cleared with Marisa as player character) before you can use other player characters there. (Incidentally, this is also how it works with unlockable non-DLC PCs.)
Partner characters added by DLCs can be used from the start with no limitations.

As for whether it's worth it or not... I think so, but as you can maybe tell from my avatar, I'm a bit biased :D If you've even a cursory interest in Touhou, definitely worth it.
For others... the spells and abilities are pretty varied. Although, even the base game easily has enough characters for 100+ hours, so if you're not sure you can just get them later.
However, at least get Sakuya DLC, it has a nice side-story and some very good dungeons. I did like Utsuho as a PC (very fun ultimate skill), and Cirno and Tenshi as a partner, so those have my recommendation... with the caveat that the fun I had with them is, of course, subjective :)
I'll be getting the whole pack. I was worried it'd make things too easy, but since it's not the case, I'd rather have the whole thing at once.
By the way, I'm entirely new to the Touhou franchise / universe, but of course I've been exposed to it. Since I enjoy Mystery Dungeon games and franchises like Neptunia and games like 100% Orange Juice, I think this is going to be right up my alley :P
