As for 2, I don't think there will be a completely customisable difficulty, although people have asked for it before. However, they will make it so you can choose that pups can/can't die from sickness, and they already have it so you can choose if you mate dies or not, so that is kind of customizible.
I feel like they said 3 won't happen, but in the new Hellroaring Mountain DLC map there is a white elk in one of the herds.
I don't know about 4, the game makers usually talk about single-player in their updates so I'm unsure what they have planned for it.
And I believe they have already said 5 won't happen, so the only pack you will belong to is the pack you create.

Quests like this were in the old game, and are not present in Anniversary Edition. I recall old dev blogs mentioning the team doesn't want to make things too grindy, which these sorts of quests impose, so I don't forsee these types of quests occuring, but I do think it'd be neat if like the baby ungulates quest some side activities will pop up on occasion to give variety while playing year to year.
2. The game currently doesn't have super deep customizability for difficulty settings like this. I know they mentioned a new upcoming option regarding pup sickness regardless of difficulty, but since each difficulty setting is more or less tuned to invoke specific experiences I'm not really sure we'll ever have a huge amount of flexibility. Especially since the game appears to be running a simulation to replicate conditions in Yellowstone National Park to a certain degree.
3. There's a leucistic elk in one of the herds in the Hellroaring Map DLC, so I don't see it as an impossibility, but I don't personally see it being something where a random chance to run into an animal with a mutation.
Currently in game pups can already grow up to have some of the modifier sliders that are based on genetic mutations (the short tail), and all black wolves in the game are melanistic. So that's kind of already existing in the game. Devs have said no albinos though, so I don't see the Saga really expanding on this much at all.
4. This is another thing we will have to wait for further development blogs for. We don't have information on how saga in multiplayer will be. Gotta be patient with this one.
5. Another thing we don't have information about. We'll have to wait and see if a devblog comes along that covers the topic, or try it out ourselves once the Saga is released.
