As long as there is two people in the game, you should be able to have pups. This is regardless of gender. There is no 'mating' depicted in the game.

If you are in story mode, first you must get enough territory, (15 hexs is enough!) and find 4 dens. Then, the game will prompt you to name your pack, and then you must chose the den, (by going up to the den), then it will prompt you to name the den, and then you will get pups! The quest will also appear on your quest marker.

You cannot have pups if you are in any other game mode however, so make sure you are in story mode! I believe that you can have pups in every map other then the classic lost river map.

Basically, make sure you are in the correct game mode (story mode) and make sure you are NOT in the classic lost river map. In story mode, it works the same as singleplayer.

Just to add, the devs have stated publicly that all wolves in a MP game with pups are canonically siblings of the litter they're raising, not parents. This prevents any weirdness especially with young kids playing the game, and is why the sex of your wolves doesn't matter.

The devblog about this explained that the game treats everyone as the genetic pool in a manner that everyone is part of a related family. Whether that means anything specific (siblings or parents or uncles/aunts or no relation that all) is up to the players participating in the game.
