How can I turn off health regeneration?

You can't. Play without using save games and it makes more sense- the regens are like extra lives, and you only get so many to finish the level. That's how I play and it's much more fun.

It does remove health regen and changes a lot of other little things that affect the gameplay to become a tad harder; Some weapons do reduced damage except when you land headshots, economy has been changed so you have to actually buy and craft stuff to survive.
Just follow the instructions in the readme;
I believe you just have to unpack it into the game directory and confirm overwrite requests.
Should you not want to play with it anymore, you can easily uninstall, tried and it worked.

What should be mentioned is that it only seems to work with the 64-bit-executable.
