No, time doesn't matter. However, you may unlock new events depending on your progress through the game.
Trees and grass don't come back naturally. You can plant grass and trees however.
Days passed don't matter. You can spend 100 days without killing the 1st boss if you want.
I have seen no evidence about the starred enemy, but there MAY be an 'for the first X days' kindness condition to not have many of them until like day 25 or something. I don't believe this either, since day 1 on my new start, a starred skeleton ate me.
Regardless, your boss shows up and glares if you are not moving fast enough to suit him. No game impact there.
while grass does not regrow, dandelions, loose wood sticks, flint/rock, and a few other such things respawn so paths with those things can have cart woes.
by the end of the game, you can regrow the grass, keep it dirt, make it black dirt or brown or mix, cover it in roman-road style tight fitting rock pattern, and possibly do other things to the ground to make it look nice.
