I mean, i started with the original RE2, barely played 1, and never felt i missed anything important. You may want to read the plot of the first, if you really feel like it.
If you're from the group of people who are in it for the story/plot - watch the previous summaries of the games. If you're about that 'gaming' with old school mechanics - start from RE1.
To answer your question: You might have some questions as to why's that or who's that, but not in a way as to not enjoy the journey that you start with RE2.
