I'd also suggest the second run after that for each, which is different in parts and worth seeing
and the extra modes are also good (but much much shorter), even if only on training mode just to see them. (for me at least training mode was a bit challenging on the hard missions)
The 2nd scenario is harder for both characters, Mr.X appear earlier and in a different in which you don't expect it, the officer's notebook is not complete (separated in two parts) and also you need a photo for the medallions (assuming you don't search for the solutions in google) and the typewritter and itembox of the main hall are not present (trust me, you will miss them a lot, specialy in early game) and there are zombies there (Marvin and two more if I remember correctly). Also, having two types of handgun will make you have less ammo, the .45 cannot be made and the 9mm can only be obtained mixing two regular gunpowders, which reduce the ammount of gunpowders for making ammo for (grenaded/shotgun shells).
In terms of the game's overall story, that's generally true, but they're different chapters of the story.
Each character's first run is very different from the other's, as they spend much of their time with different people, sometimes in different locations.
