Try to grab key items before consumables, but remember you can throw out any item except weapons and key items.
You can also detach weapon parts. Not sure if you can throw them out.
Don't forget to pick up every inventory upgrade. Leave healing items where they are and only pick em up when you need them or right before a boss fight.
Since you are going on assisted, you should also try getting the "no healing" achievement, since on assisted you get healed to 50% of your health after you take a hit. Doing both of these at once can be a bit of a pain, but if you know what you're doing you won't really have anyh issues. That is, of course, if you even care about achievements. If you are just doing this run because you feel like it, then you can ignore that part.
Remember you can't discard guns, or grab any bonus weapons either. Claire is probably the better choice, you could probably forgo the stock and suppressor on both the grenade launcher and SMG just this once for the challenge run for more space, although I don't think you'll have a problem with the space personally.
You'll need to be confident in your health supplies though, no real easy stockpiling, which is the hardest part. I kinda recommend doing it alongside a speedrun, so you spend less time at risk of taking damage. Since people are pretty adept at beating the game in an hour without touching the item box, it shouldn't be too tricky.
The worst part I came across was the large gear for the clock tower, because I also had a heart key and a diamond key I couldn't discard at the time, but you definitely get enough inventory space to bypass it, though.
It's not as hard as you think anyway, once you stock up on the hip pouches. You can claim two in the first five minutes of the game as well.

Yes, for Leon, the Diamond Key opens a room in the Firing Range (Red Herb, Roll Film) and the Linen Closet on the west side, second floor of the RPD (Portable Safe, Gunpowder).
Claire requires it to gain access to the elevator controls for the Chief's Office. Other than that it does the same thing for her that it did Leon.
You will lose access to a Hip Pouch by skipping this key.
No, infinite weapons are inside an item box
