this game takes place during the peloponesian war which lasted from 431–404 BC, alexander was born in 356 BC.
that alexios is immortal does not matter, the game is not set in a time in which alexander was alive.
Yea, in order to add what OP asks they need to change the game world and it won't happen i guess
BC is backwards, 431-404 BC happened first then 356 BC happened second..
So Thermopylae happened first before Alexander.

its not that alexander would not be possible with alexios, it is that the map in which the peloponesian war is going on is not really usable for such a dlc and therefore it would require its own map of greece alongise the map expansions to cestiphon or norhtern india.

of course it is backwards, but the map does not go further in time, the peloponesian war is going on all the time at the map, the game does not go further in time than the duration of the peloponesian war.
and what exactly has the battle of the thermopylae to do with that? that battle already happened before the peloponesian war and is part of the prologue.
a alexander dlc would require its own map of greece alongside some map expansions, if something like that happens then it would be more likely that it would come as a standalone game rather than a dlc i think.
