And can i enable the dawnguard dlc again whenever i want ?

Nvm, it is indeed only destroyed with Dawnguard. There's one NPC there that train Restoration and will die if you enable Dawnguard, though she's not a Master Trainer so she's not that important.
In addition, Dawnguard introduces another master trainer in Restoration (who you find and bring back to the Dawnguard, if you join the Dawnguard) who also sells powerful anti-undead spells (they function like fireball and flame cloak respectively, but only hurt vampires and other undead, not living creatures).
While Keeper Carcette also would cure your character of diseases for free if you asked, you can get the same effect from any shrine, AND get a buff from that shrine as well.
So basically, you don't lose anything particularly unique or super-useful when the Hall of Vigilants is destroyed, so no need to worry about it! :)
But no, you did not miss anything crucial, unless you're a 100 percent completionist. In that case you'd have to clear the hall before it gets destroyed (which means murdering non-hostile NPCs: All inside and two outside, so the location shows as cleared, it also means postponing leveling until you raised pickpocketing really really high - so not worth the effort if you have Dawnguard installed).
Personally, I can not even seem to clear it although I killed everyone. So maybe it is bugged or was patched later to not be clearable anymore.
(And yes, I know the thread is two years old, but there might be other people wondering, so they can be informed.)
