Can you join all the guilds?
Or does joining one conflicts with another?

You can but it might not be very lore-friendly.

True - and role-playing within reasonable lore/realism limits is important for me.

Though there are ways to prevent yourself from joining certain factions, joining other factions isn't one of them. Skyim doesn't really have any "guilds", as such...

The exception would be the whole Stormcloaks / Imperials thing, though even then you get the option to switch sides early on.

There aren't any factions that are "essential" to join, but do keep the Stones of Barenziah[www.uesp.net] in mind.

But boy; that's a long, involved quest! How long does a normal, reasonably-thorough gameplay take? I've played 36 hours, and I am only level 26, and I feel I have not even done a quarter of the game?!

Of course, this game seems to have less re-playability than other similar high-end RPGs, since the classes are rather limited - so I guess it evens out.

Heard it takes around 250-300 hours to 100% this game.
Also, I can't agree about the replaiabilty part. I think I've started a new game 20+ times now. A lot of people did aswell.

The lack of replayable classes is the big thing for me - as I exclusively play melee damage classes. So at best I can go dual wield DPS and 2H DPS for 2 games. (Compare this to the seemingly infinite variability of class or multi-class configurations in NWN 2.) But I understand people replay for different reasons.

The quest I referred to is really for the "complete everything with one character"-sort of player, as opposed to the "I'm only going to do quests of a certain type"-sort of player. It's quite possible to complete it without "trying", but without a guide, it takes a fair while to accomplish.

In truth, Skyrim's classes only exist to the point you can imagine them - this is true of other TES games as well, but Skyrim makes it even more "obvious". It's quite possible to create a character who is the master of everything, and in the long run, the "jack of all trades" beats all.

you are Dragonborn you can do whatever you want and whoever disagrees with you you can shatter him into tiny pieces with your Thu'um

but seriously there are no restrictions to what you can do you can be a thief and a murderer who gives charity and raises children

there are no factions actualy in Skyrim .. these "guilds" are just another quest lines you can do now and then completely ignore later

and some people say its not lore friendly ... bollocks
you can be a two faced person like Paul De Pleur from Far Cry 4 and it won't break immersion

Also its really beneficial to not be a roleplay freak and actualy do these quest chains

EDIT:
dabble in everything in your first playthrough to check what this game has to offer
instead of missing out

fully roleplay and restrain yourself later in second playthrough

Well it also depends on the mods your playing with, for instance skyre and perma have so many perks you can't do it all.

well if you limit yourself to not being evil or roguish then your limiting yourself not the game just saying

Mods give it loads more replayability. I'm just doing a playthrough now with a group of followers, they do all the fighting and I'm being a sort of healer and keeping them alive. Good fun.
FYI You will need to join the College of Winterhold to complete the main questline so staying "true" to any ohter guild is impossible.

Pretty sure you don't have to join them. You can show the guard chick your Thu'um and she'll let you in. The college I mean. Nothing sexual. Maybe.

no you dont .. Urag will help you regardless of being a member of the college or not and Farada will let you in if you demonstrate power of the voice to her
