On the road you get crazy random events.
If you click on the assassin or the note (probably need to drop it first) in the console you can compare the first two characters to your mod list - it'll be in hex so you'll need to compare to whichever of the two (in MO it's priority and mod index, with mod index being the relevant one, in NMM it's the other way round with slightly different names, I think?)
Nope, there's three or four instances in the vanilla game when someone will attack you with a 'contract' in their inventory after you kill them:
Hired Thugs - theft. If you steal something (regardless whether you're caught or not) the owner (or someone linked to the location - e.g. in Dragonsreach it's, like, the cook or something) will hire thugs to 'teach you a lesson. They carry a note from the person who hired them[www.uesp.net]
Hired Thugs - murder. Pretty much the same deal except this is for having murdered someone. They carry a note from your victim's relative[www.uesp.net]
Normally only one set of Hired Thugs will come after you per playthrough.
Dark Brotherhood Assassin. These are entirely random after level 5 (they are NOT in response to ANYTHING you have done), but should stop once you've completed either With Friends Like These... or Destroy the Dark Brotherhood (i.e. after you've either joined them or beaten them.) They carry a note signed by Astrid[www.uesp.net]
Thalmor Death Squad. Can happen pretty much any time after level 8, apparently, although I've been lucky and usually don't encounter them until late enough in the game that it makes sense. They carry an Execution Order[www.uesp.net]
As you can see and as OP said, this is a completely different note.
