Thats a scripted event. Happens when tou first enter Markarth.
You need to follow up on the murder to get the quest. Talk to the victims family and/or get the message from the killers body.
Nope, in normal gameplay (without mods) you don't find any message on the body, the victim's family are in Cyrodiil (no mod that changes that AFAIK), and you are normally force-greeted by Eltrys who comes to very subtly give you a note that you 'dropped'. I think Immersive Citizens may also change this, btw (because I've been playing with that, forgot to install TCIY and still didn't have him force greet me, IIRC.)
Could be I haven't played that part for years. But didn't the victim have a husband and son you can talk to afterwards? And I do remember that 'drop' part, but as he said he ignored and left the quests.
One can always use console to start the quest.
Nope - if you save her you can talk to her, otherwise it's just a case of looting her corpse for her room key/picking the lock to her room. You may be thinking of the optional dialogue options that the female jeweller and the guy who sells meat in the marketplace have? (though yeah, it all does tend to blur after a while!)

Really not advisable to use the console for this kind of thing unless *nothing* else is working and it's essential to your playthrough that you do the quest.
It messes also whit the events in Solitude. It is made for stopping couriers to talk and stop you, but is stops quest like the one you have issues whit.
