Since I don't know where Misfortune sprinkled sparkle in your gameplay, I can't say which one she missed. You may wish to consult a guide to all the sparkle places. Here's one that may help: https://steamcommunity.com/sharedfiles/filedetails/?id=1872361778.
You can either play with the puppy or free him and still get sparkle on all the required spots to get the additional content in the game. The sparkle place changes depending on which choice you make regarding the puppy.
And as far as doing another full run, that would depend on whether or not you have a save game prior to the spot Misfortune missed spreading sparkle on. If it is the spot that is near the trash on the beach, and the dock to George's house that you missed, then you can use a save game from that point on (if you sprinkled sparkle on all the required spots up to that point before making the save game) to the end of the game and sprinkle sparkle on all the rest of the required spots again. If you don't have a save game there, then yes, you'll have to do another full run. You must sprinkle sparkle on all the required places in a single playthrough of the game.
You might want to look at this reply of mine on another topic for some save game files that might shorten your replay time if all you want to do is see the ending you get for sprinkling sparkle in all the right places: https://steamcommunity.com/app/714120/discussions/0/4078523564600309427/#c4078523564601058771. Using one of those files and playing to the end will get you all the sparkle spots up to that point, and you need to replay to the end of the game from there.

Thanks for replying. Ah yes, the switch version just became available. Thanks for your support there! I haven't seen the Switch version yet, but I'm sure it plays the same as on the PC. Of course you cannot use the computer saves on the Switch, so that didn't help you at all.
You will need to play either all the way through from beginning to end again on the Switch, or from when you have a save slot just before the decision point where you either can choose to "Play with the puppy" or "Set it free".
Since, in your first playthrough, you chose to "Play with the puppy" and got the ball, then the location where you sprinkle sparkle is not at the beach with the trash and the dock to George's house.
Instead something bad happened to the puppy where you made the decision to "Play with the puppy", and you need to sprinkle sparkle there. And you should see and hear the "sprinkle sparkle" effect there too, just as you see and hear those effects at all the other "sprinkle sparkle" spots.
And you'll need to play throught the rest of the game and again sprinkle sparkle at all the remaining spots (as shown in the guide I referred you to in my first post) and that you did in your earlier gameplay. All the glitter spots must have sparkle again spread by Misfortune in a single playthrough in order for you to see the additional ending.
If you do play again and choose to "Set the puppy free" instead in that playthrough, then the beach with the trash and dock IS the place to sprinkle sparkle and you'll see and hear the effects that happen at all the other "sprinkle sparkle" spot there instead of where you first made the decision about the dog.

Oops I got confused! The Switch version of Little Misfortune has actually been out for some time. It's the Fran Bow Switch version that just got released last Friday. And I've been busy answering posts in the Fran Bow forum here on Steam since then, and forgot that I was dealing with Little Misfortune with your post.
Anyway, thanks for the double support on Switch! It's greatly appreciated! And I hope you enjoy playing Fran Bow as well.
