Do I need to complete all these quests in a single run?

I have beat the game twice now, and I missed that stupid 5-statue elevator quest both times. The first time, I didn't realize the level would just end after I beat the boss. The second time, I completely freakin' forgot that the level just ends after the boss.

yep all quests must be done in an single run think i beat it two or three times to get everything

Can confirm, just for anyone else curious about this achievement:

You do have to finish all the quests in a single run, but you don't have to beat the game. Not that it matters, since the last quest is right outside the Big Boss' door.
