How do I get the the chests or through the vines

I think you are talking about the puzzle chests.

If you look at the walls inside the tile with the chest you will see a grid with marked locations. You have to place your characters on the marked locations. There should be a red marker on the wall grids to help you with the proper alignment.

once you have the characters positioned the vines will fall?

I have players in position and vines don't drop

You have to align them in the correct position compared to the puzzle itself.
Its not very intuitive, but after a while you will get the hang of it.
