Yes, managers do get "tired" if you they do too many shifts in a row them and this effects their "Influence" stat. Basically for (almost) each shift their effectiveness drops by 1. So switch in a less-tired manager when your main one has worked four/five shifts in a row.
That said, most of what I've read said the change is negligible and all it does is lead on slightly less money. If you're like me and you don't need much cash, you can just leave the tired managers in and not worry about a little money lost.
If playing as Majima, it's even easier since you can buy the Slime Gun shotgun from the Crafting place right from the start. Get 4 or 5 of those and use them on Shakedown for quicker fights...then head back to the crafting place to "repair" them and refill the ammo for the next encounter. I think the weapon is unlocked for Kiryu at some time too but I'm not sure when.
So, it's not that big a deal if you ignore managers. At the same it's pretty easy to switch a manager out if you see a negative number. Even better, once a manager is switched out, their meter resets so you can just assign them to another district and they're fresh and ready to go again.
In the end, if I really want to make cash as Kiryu, I'll lose to Mr Shakedown then use Kiryu's multiplier to win 150% of it back. Much easier than worrying about small losses in Real Estate anyway.
