They'd better keep them in sequels, because while Nero on his own is fun, he's not the reason why I replay DMC games.
I think this is a great chance to mix and re-create all the best in the combat mechanics of all the characters.
For example, a character who fights in a style similar to Vergil, but with the ability to switch styles with different priorities in battle.
Dante's combat mechanics are constrained by a weapon switching system.
The second new character can have multiple weapon modes, like King Cerberus.
Movement with each weapon can show different fighting styles more strongly: damage, attack and evasion (Balrog), weapons that parry attacks, etc.
Switch weapons bind to D-pad for instant access to any weapon.
The series has huge potential for evolution and new ideas)

I don't understand in what universe that this is their perfect ending.
First we need a game where both Sons are playable in the main story before the series should call it quits. Especially when there are still unanswered questions.
But more than that, the plot seems prime for it. Dante and Vergil fighting Demons as they're stuck in the underworld and have to make their way back, Nero on the surface with Lady and Trish, I feel like everything's set up perfectly for both of these groups to have a campaign that can make good use of a co-op experience.
It feels like the only place where DMC gameplay can evolve to. Like I said, DMC5 seems like Peak DMC, peak combat, peak gameplay, peak style. Co-op would be it growing 'sideways' (or, to put it another way, growing in a direction that it previously wasn't growing in), and I think that's where the most growth could be found for the series at this point.
If its optional... fine, I won't use it, but I don't want anyone to help me kill demons in the actual game. I want to earn those ranks you know?
However, to have co-op in every mission would be an obnoxious feature. As an unlockable setting for beating normal bloody palace, a co-op bloody palace with tougher variants of enemies, maybe even returning enemies who don't appear in the main game sounds fantastic!
I think the most obvious answer is that with both the Son's of Sparda carving away at hell itself, and the weakening barrier between worlds, there will be a mass diaspora of demon-kind from various circles of hell. It wouldn't be surprising if reality began to stop making sense and we started seeing Earth fracturing between the human and demon realms like we saw in the reboot. Entire cities have already been leveled by demonic incursions. Nero and the earth-side crew will have their hands full with no shortage of jobs to take, and new rivals competing for business. Now is a ripe time for new characters and rivalries to be introduced.
I think a prominent plot point will be repairing the seal between the human and demon realms, and whatever sacrifice it will require, since we've already seen some the herculean efforts Sparda went through to seal the worlds off the first time.
