In the Devil May Cry novel Gilver said that if he cut out Dante's heart, he wouldn't be able to recover, no matter how much demon blood runs through his veins.
I have to point out that the novel is semi-canon, but I guess this statement has a hint of truth. Hideki Kamiya also said that the heart is the most important, so this might be the case, even more that Vergil was "dead" when Mundus removed his heart.

Moreover, we've seen many times that these two were stabbed in the chest with a sword and nothing serious happened to them, but "cutting out" and "stabbing" are probably two different stuffs and maybe they can't regenerate after that, but who really knows, with them everything is possible~

I don't know why Nero is said by some people to be the most powerful after one fight, which Vergil gave him a really huge advantage and didn't want to fight him. I agree, Dante and Vergil're more powerful than Nero, still according to all their characteristics. I still think Nero'd have no chance if he fought the twins from Dmc3. Nero has the power, but considering what these two can do. They're also more experienced now and it's clear that Nero still has no chances if they fight him seriously. Nero may have more raw power, but that's not the most important thing, because if someone doesn't fight with his head, we have the example of the wounded Vergil fighting Mundus. Strength'll do him no good. Nero is a hothead, that's why he'll never reach the Level of Vergil or Dante. Maybe if he changes his approach which I doubt, because each character has its own traits. Vergil's always calm and cool, Dante's coocky and goofs around, Nero's hothead and combative :PatchouliKnowledge:
Compare Nero in DMC4 to Dante in DMC3. They are of a similar age.
Dante has natural talent with weapons and is extremely agile and skilled even before his DT awakens. Nero on the other hand is a little slower and less skillful, but he is way stronger than Dante was shown to be. He can toss monsters around with ease, even huge ones like Belial. And he performed some insane feats like tossing around the giant Sparda statue sword and crushing the savior's face bare-handed (kind of)
Nero's age: 16-17 (Dmc4), 22-23 (Dmc5)
Dante, Vergil's age: 19 (Dmc3), 41-42 (Dmc5)

Nero was between and is still between Vergil's age from DMC3. Yet he is still no match for them, Nero's raw power can be bigger but it doesn't make him better fighter than brothers were from Dmc3, and I said what traits are resposnible for it. He still can be defeated by them if it comes to fight "live or die.
A serious fight with Vergil would end with Nero's death, because Vergil has control over himself, also super speed, power, strength, experience, especially since Vergil fought constantly in hell. Although in this circumstance it was obvious that Vergil couldn't cope with his emotions during the fight with Nero in mission 20. He gave Nero a head start.
