I don't really care too much which heroes I use. I equip all of my rings in order to earn more runestones while playing. I don't have the last four alternate characters, but of the first three I would prefer Defender, Ranger, and Priest. I like the Defender because of his Decisive Cleave skill, this might be either my first or second favourite skill. I like the Ranger for her aforementioned boss killing skills, and I don't need the Huntress's speed since I use a Summon Friend book all the time. The Priest is fine for heals, shields, MPS bonus, and he frees up the shield points that the Defender would normally use.
As for the other character's skills:
Thief - Max out Shakedown and Share. No other skills matter to me.
Fiery Fox - One of the best characters in the game in my opinion. I'll put a single point in Spirit of the Fox, max out Rage and Rampage, and split the rest in Regeneration and Ritual. His Spirit of Fox battle ability and the Defender's Decisive Cleave were the two abilities that helped me beat level 3,000 for the first time.
Enchantress - I'll max out Thunderbolt for the possibility to hit all enemies and get through my run a little bit faster. The rest goes to Discharge and Ice Rain.
Guerrilla - Another pretty good character. I'll max out Turret, put five points in Aim, and the rest get split between Barrage and Mines.
